In short

An area around the car that the driver cannot see using mirrors alone; requires an over-the-shoulder check.

A blind spot is a region around the car not visible in any mirror. For a typical sedan, the rear corners on each side are the main blind spots.

US examiners specifically watch for over-the-shoulder blind-spot checks before lane changes. Missing one is a top-scored fault.
